This continuing education course will cover the clinically significant drug–drug interactions (DDIs) associated with oral anticancer therapies. The presentation will cover potential adverse effects of DDIs and examine key interaction pathways, including those involving CYP3A4 inhibitors and inducers, gastric acid–suppressing agents, and medications that prolong the QT interval. Strategies for identifying, preventing, and managing these interactions will be discussed to help ensure patient safety and maintain optimal therapeutic efficacy.

At the end of this session, learners should be able to:

  • Identify oral oncology agents with high drug–drug interaction (DDI) risk and describe the key pharmacokinetic and pharmacodynamic mechanisms that contribute to potential interaction.
  • Explain how drug interactions involving CYP3A4 inhibitors and inducers influence clinical outcomes.
  • Describe the ways acid-suppressing agents interact with oral chemotherapy therapies and summarize considerations for management.
  • Explain how QT-prolonging agents interact with oral chemotherapy and outline key principles to monitor and prevent QTc prolongation.
